Miami is the most convenient airport if traveling from Europe. Otherwise, Tampa, Fort Myers or Naples airports may do the job. Orlando will have more and cheaper lights and car hire is more accessible there than Miami (where you have to take a shuttle bus from the terminal to the various car rental premises), but then from Orlando you have a 3.5 to 4 hour drive to Naples. Worth thinking about if you've been traveling for 12 hours plus already.
If traveling via Miami, just go west on I-75 (the green road on the map above) for about 2 hours and you hit Naples. It's a nice drive along 'Alligator Alley' which has been cut right through Big Cypress Preserve. About half way across on the right there is a gas station, store and eatery (with toilets) - the only one between Miami and Naples. Have a look at the pond next to the off ramp - alligators are often sat there in the hopes of an easy meal. Don't worry, theres a fence to protect the alligators.
Once into Naples, you can either get onto US 41 (Tamiami Trail), go north and 99th Avenue North is signposted off to your left or stay on I-75 and turn left at the junction of Immokalee Road (junction 111), left again onto US 41 and 99th Avenue North is on your right. Couldn't be easier! The link below gives a clearer picture.

Local Amenities:
Virtually everything you could possibly need will be found somewhere along US41(Tamiami Trail). Turn right out of the driveway, carry on until you hit a main road (that's US41 - Tamiami Trail). Turn left or right - you'll find a shopping centre in either direction. If you turn right, you'll find the Pavilion Shopping Center (at the junction of Tamiami Trail and Vanderbilt Beach Road) has everything you need - supermarket (Publix), banks, bars, restaurants, gas station and even a cinema. If you turn left, there's more shopping etc on the north side of the junction of Tamiami Trail and 111th Avenue North.
Restaurants:
Carry on past the Pavilion shopping centre (south on US41 - Tamiami Trail) and you'll find numerous restaurants lining the road. I can recommend brunch at Perkins (great pancakes and hash browns), a succulent steak at the Texas Long Horn, the Outback (Australian theme) or the Cheesecake Factory (more than just chesecake). For that special occassion, Fredericks is quite 'up-market' and serves excellent food which is 'reassuringly expensive' - whatever that means.
Beaches:
If you go west at the junction of 111th Avenue North and Vanderbilt Drive (Bluebill Avenue), you'll find a gorgeous beach and nature reserve - Delnor-Wiggins Pass Sate Park - with barbecue areas, toilets etc. No alcohol permitted and parking is $5. Entry is free if you're on foot.
There is another public beach at the end of Vanderbilt Beach Road - at the southern end of Gulf Shore Drive. Parking is on the street - meter charges apply.
Both these beaches are about 15 minutes on foot from Sam 2.
If you'd like to enjoy a cocktail whilst watching the sun set, the La Playa Beach & Golf Resort (Gulf Shore Drive) is open to non-residents and has a very pleasant bar fronting the beach. The foods pretty good too.
